Interactions between enamel matrix derivative (EMD) and human periodontal ligament (PDL) fibroblasts were investigated by culturing fibroblasts in the presence of fluorescently labeled EMD and measuring the cellular uptake of EMD by immunohistochemistry and laser scanning microscopy. The human PDL fibroblasts internalized EMD, giving vesicle-like structures in the cytoplasm. The internalized material comprised mainly the 20 kDa amelogenin component, which was subsequently processed to a cumulative 5 kDa component.
